目的 通过向培养液中添加不同浓度的硫酸亚铁和三氯化铁研究其对盐藻生长的影响,及盐藻对不同价态铁的吸收机制。方法 紫外分光光度计检测其对盐藻生长的影响,原子吸收检测盐藻胞外溶解态、吸附态和胞内吸收态铁的富集规律,并用红外光谱检测盐藻富集前后红外吸收的变化。结果 当硫酸亚铁的浓度为12.5 μmol.L-1、三氯化铁的浓度为25 μmol.L-1时,盐藻生长较旺盛。随着培养周期的延长,胞外溶解态铁逐渐降低,膜吸附态和胞内吸收态铁呈现增长趋势,整个培养周期,铁的总量保持不变。另外,铁离子的添加并未对盐藻细胞分子官能团产生伤害。结论 当硫酸亚铁浓度为12.5 μmol.L-1时,盐藻具有最好的生长状态,Fe2 是盐藻的主要吸收形式,盐藻对铁的吸收是一个循序渐进的过程。 |

Objective To study the absorption mechanism of Dunaliella salina under different iron concentrations and valence. Methods The growth curve of D. salina was measured by ultraviolet spectrophotometer. Dissolved, adsorption and absorption state of iron were determined by atomic absorption spectrometry, and the change of infrared absorption spectra were analyzed before and after iron bio-enrichment. Results When D. salina exposed to 12.5 μmol.L-1 Fe2 or 25 μmol.L-1 Fe3 , it had the highest growth rate and absorption efficiency. As culture cycle went on, extracellular solution iron decreased constantly, adsorption and intracellular absorbtion iron showed a rise. The total of iron remained unchanged during the whole culture cycle. Addition of iron to Dunaliella salina did not harm to the functional groups. Conclusion D. salina grew best in the medium with 12.5 μmol.L-1 FeSO4. Ferrous is the main absorbed form of D. salina, and the absorption of iron is a gradual process. |
