      目的 研究鲨鱼皮胶原(Shark skin collagen,SSC)海绵的体外抗氧化功效并研究其体内促伤口愈合作用。方法 采用醋酸溶胀法分离酸溶性Ⅰ型胶原,真空冷冻干燥技术制备SSC海绵,化学分光光度法检测其体外抗氧化活性;采用SD 大鼠背部创伤模型,检测SSC海绵的促愈合作用,HE染色观察组织病理学变化,免疫组织化学染色检测愈合过程中CD31,EGF和TGF-β表达变化。结果 20 mg/mL SSC溶液对HO? 具有显著的清除作用,清除率为84%±0.021。术后第3、5天,SSC 海绵处理组愈合率分别为52.1%±0.057、57.1%±0.080,显著优于纱布(gauze,GZ)组的15.9%±0.083、28.4%±0.066 (p<0.01)和壳聚糖(chitosan,CS)组的22.2%±0.095、36.5%±0.033(p<0.05)。组织病理HE染色显示,SSC海绵能在创伤愈合早期显著促进毛细血管生成,新生肉芽组织形成,促进愈合上皮生成,加速创面愈合。免疫组化检测显示,术后第3天,SSC 海绵处理组CD31表达数为38±4.73,显著高于CS组的27±3.06和GZ组的26±3.61(P<0.05);SSC海绵处理组TGF-β表达率为20.1%±3.17,显著高于GZ组的6.7%±0.739和CS组的10.7%±0.502(p<0.05); SSC海绵处理组和CS处理组EGF表达率分别为12.1%±0.447和12.2%±1.91,显著高于GZ处理组的9.3%±1.23( p<0.05)。结论 SSC具有显著的清除羟自由基功效,SSC海绵能够显著促进创伤愈合早期CD31、TGF-β和EGF的高表达,进而促进早期新生血管生成和肉芽组织的形成,加快伤口愈合过程。
English Summary:
      Objective To study the effect of antioxidative activity in vitro and promoting wound healing in vivo of shark skin collagen (SSC) sponge. Methods Acid soluble SSC was isolated by swelling with acetic acid, SSC sponge was obtained by freeze-drying. Antioxidative activity of the SSC was detected by chemical UV test. Rat Back Wound model was used to evaluate the effect of SSC sponge on wound healing. Histopathologic examination of the wound was studied by HE stain. The expression of CD 31, EGF and TGF-β was determined by immunohistochemistry. Results DPPH? , HO? scavenging ratio and ferric-reducing power of 20mg/mL SSC were 18%±0.016, 84%±0.021 and 0.11±0.002,respectively. On the 3rd day and the 5th day after operation, the healing rates of SSC sponge treated groups were 52.1%, 57.1%, respectively, and they increased significantly compared with the GZ-treated groups of 15.9%, 28.4% (p<0.01) and the CS-treated groups of 22.2%, 36.5% (p<0.05). SSC sponge promoted the formation of blood capillary, akaryocyte and granulation in the initial stage. On the 3rd day, the expression of CD31 in SSC sponge group was 38, and increased significantly compared with the CS group of 27 and the GZ group of 26 (p<0.05). The expression ratio of TGF-β in SSC sponge-treated group was 20.1%, and increased significantly compared with the GZ group of 6.7% and the CS group of 10.7% (p<0.05). On the 3rd day, the expression ratio of EGF in SSC sponge group and CS group were 12.1% and 12.2%, respectively, and increased significantly compared with the GZ group of 9.3% (p<0.05). Conclusion SSC has strong effect on the elimination of HO? . SSC sponge could promote the expression of CD31, TGF-β and EGF in initial stage of wound healing. It also significantly promote the formation of vascellum and granulation to accelerate wound healing.
