目的 运用基于标准浸膏特征谱图-指标成分-药效的多指标综合评分法确定经方牡蛎散的最佳提取工艺。方法 利用HPLC法建立牡蛎散标准浸膏特征图谱,并建立指标成分毛蕊异黄酮苷的含量测定方法,采用小鼠体外脾淋巴细胞增殖实验评价浸膏的免疫活性;以与标准浸膏特征图谱的相似度、毛蕊异黄酮苷提取率和体外脾细胞增殖率为指标,采用L9(34)正交试验设计法对牡蛎散进行提取工艺优化,运用多指标综合评分法确定最优提取工艺参数。结果 牡蛎散的最佳提取工艺为:药材加8倍量水,浸泡1 h,提取2 h,提取2次。结论 优选的提取工艺合理可靠,适合工业化生产;多指标设计探索将化学与生物活性相结合、特征图谱与指标成分相结合,为经方提取工艺优化研究提供了方法学参考。 |

Objective To optimize the extraction process conditions of Muli San by Multi-index “HPLC Characteristic Spectra of Standard Extract-Index Compotent-Pharmacodymamics”. Methods To establish the characteristic spectra of standard extract and a method for determination of calycosin-7-glucoside by HPLC, splenic lymphocyte proliferation was used for evaluation immune activities. L9(34)orthogonal design with Multi-index were used to optimize the extraction process conditions for Muli San. Results Optimum extraction conditions were as follows: extracted two times with 8 times the amount of water for 2 h every time. Conclusion The optimized extraction process is reasonable and reliable for industrial production. Multiple indexes combined chemical and biological methods、characteristic spectra and index compotent, which can provide methodology references for extraction process optimization study of Classical Famous Prescription. |
