      目的 研究天然鱼肝油与维生素AD滴剂对去卵巢大鼠骨质疏松症的改善作用。方法 比较分析天然鱼肝油与维生素AD滴剂的脂肪酸组成差异;对雌性SD大鼠进行双侧卵巢切除术,建立骨质疏松模型,术后4周随机分为假手术组(生理盐水)、模型组(生理盐水)、维生素AD滴剂组(0.25 g·kg-1·bw-1)、鱼肝油低、中、高剂量组(0.25、0.5、1.5 g·kg-1·bw-1)。连续灌胃12周,称量体重及各脏器,计算脏器指数,分析尿液骨代谢指标(钙、磷)、血清骨代谢指标(钙、磷、肌酐、骨钙素、降钙素、碱性磷酸酶、雌二醇)、骨物理指标、骨钙含量、骨密度并对肝脏组织进行HE染色观察,考察天然鱼肝油和维生素AD滴剂对大鼠骨质疏松症的改善效果。结果 天然鱼肝油的脂肪酸种类更加丰富,并含有EPA和DHA两种n-3多不饱和脂肪酸;天然鱼肝油不会对肥胖进展有促进作用,可在一定程度上恢复子宫形态,防止钙、磷流失(P<0.05);两种物质均可使血清中钙、磷水平恢复并降低碱性磷酸酶水平(P<0.05),鱼肝油可降低骨钙素水平(P<0.05);两种受试物均显著促进骨钙含量增加(P<0.05),并提高骨密度(P<0.05),证明两者可减少骨量丢失,对改善骨质疏松有良好效果;肝脏病理学观察显示相较于维生素AD滴剂,天然鱼肝油更能抑制骨质疏松导致的肝脏脂肪积累。结论 天然鱼肝油和维生素AD滴剂均具有改善去卵巢大鼠骨质疏松的作用,前者的改善效果更加理想。
English Summary:
      Abstract: Objective This study aimed to investigate the ameliorative effect of raw cod liver oil and vitamin AD drops on osteoporosis in ovariectomized rats. Methods The fatty acid composition of raw cod liver oil and vitamin AD drops was analyzed and compared. Female SD rats were ovariectomized to establish an osteoporotic model, then randomly assigned into the sham-operated group(normal saline), model group(normal saline), vitamin AD drops group(0.25 g·kg-1·bw-1), cod liver oil low, medium, and high dose groups(0.25、0.5、1.5 g·kg-1·bw-1) four weeks later. After 12 weeks of continuous gavage, various parameters including body weight, organ/body weight ratios, urine bone metabolism indexes (calcium and phosphorus), serum bone metabolism indexes (calcium, phosphorus, creatinine, osteocalcin, calcitonin, alkaline phosphatase, estradiol), bone physical indexes, bone calcium content and bone mineral density were analyzed; the liver tissues were observed by HE staining to investigate the ameliorative effect of raw cod liver oil and vitamin AD drops on osteoporosis in rats. Results Compared with vitamin AD drops group, raw cod liver oil contained a more abundant variety of fatty acids including two n-3 polyunsaturated fatty acids EPA and DHA; it did not promote obesity progression but could restore uterine morphology to some extent while preventing loss of calcium and phosphorus(P<0.05). Both substances restored levels of serum calcium and phosphorus while reducing alkaline phosphatase level(P<0.05); additionally, cod liver oil reduced serum osteocalcin level(P<0.05). The bone calcium content/mineral density significantly increased in both groups treated with either substance(P<0.05), indicating improvement in osteoporosis by reducing bone loss. The histopathological examination of the liver revealed that raw cod liver oil exhibited superior efficacy compared to vitamin AD drops in suppressing hepatic lipid accumulation induced by osteoporosis. Conclusion Both raw cod liver oil and vitamin AD drops demonstrated potential for ameliorating osteoporosis in ovariectomized rats, with the former exhibiting a more favorable therapeutic effect.
