| 应用ＤＮＡ重组技术将ａ型重组人肿瘤坏死因子衍生物（ｒｅｃｏｍｂｉｎａｎｔｈｕｍａｎｔｕｍｏｒｎｅｃｒｏｓｉｓｆａｃｔｏｒａ，ｒｈＴＮＦａ）ｃＤＮＡ插到穿梭质粒ＰＤＣ－８上，构建成穿梭表达载体ＰＤＣ－ＴＮＦ，转入鱼腥藻ＰＣＣ－７１２２０中进行表达。 |

| Recombinant human tumor necrosis factor a analog (rhTNF-a)cDNA was linked with the shuttle vector PDC-8 and resulted in expressing vecto PDC-TNF. PDC-TNF was transfered into Anabaena sp.PCC7120. After the culture,we found the expression of the rhTNF-a in Anabaena sp.PCC7120 was about 15% by SDS-PAGE and wester blotting. rhTNF-a was purified to about 90% purity by ultrasonication and ion-exchange chromatographic procedures. A molecular mass of approximately 17. 3kD and a specific activity of 2. 1 X 106 IU/mg were found. |
