      海带经酶解 ,季胺盐及乙醇分级沉淀 ,离子交换 ,凝胶层析等处理 ,从中逐级分离到 3种多糖 :粗多糖 ,岩藻半乳多糖硫酸酯 ( FGS) ,FGS的高纯组分之一 F4 。经口 FGS,给药一次 ,对正常小鼠及四氧嘧啶致高血糖小鼠降糖作用不明显 ,但长期给药 ( 2 0 d) ,剂量在每天 80 0 mg· kg-1时 ,粗多糖 ,FGS分别可使四氧嘧啶致糖尿病小鼠的血糖水平降到 82 .3% ( P<0 .0 5)和 76.2 % ( P<0 .0 1)。腹注 F4 ,剂量在 50 0 mg· kg-1,对正常小鼠及四氧嘧啶致高血糖小鼠 ,分别在给药 3h与 7h,使血糖水平降到 68.5%和 52 .8% ,且可在 2 4 h内维持降糖作用。急毒实验证明 :FGS对小鼠无毒害作用。
English Summary:
      Three polysaccharides: rude polysaccharide, fucoidan galactosan sulfate (FGS) and the high pure fraction (F 4) of the FGS were separated gradually from Laminaria Japonica by enzymolysis, quaternary ammonium salt and alcohol fractional preciptation, ino exchange and gel filtration chromatography. When poured p.o. to normal mice and alloxan induced hyperglycemic mice one time, FGS did not exhibit remarkable hypoglycemic effect. But if poured p.o. for a long time (20d), the rude polysaccharide and FGS could the blood glucose level of the alloxan induced diabetic mice to 82.3% (P<0.05) and 76.2% (P< 0.01 ) at the dose of 800mg/kg.d. And at 3h and 7h after p.o. administration, F 4 could reduce the blood glucose level of normal mice and alloxan induced mice to 68.5% and 52.8% at the dose of 50mg/kg and the activity could been maintained within 24h. Meanwhile FGS did not poisonous to mice on p.o. administration.
