摘要:目的 研究方格星虫多糖(SNPS)对低剂量中子辐射损伤大鼠的保护作用。方法 SNPS 3个剂量组70,140和280 mg kg-1。补充何种动物、数量、性别、级别,重量,SD大鼠,50只,雄性,清洁级,180-200 g,每天于照射前经口给予受试药,连续38 d。除正常对照组外,其余各组每天照射22 h,放射源为252Cf中子源,吸收剂量率为0.085 mGy h-1,累积吸收剂量为71.06 mGy。在照射第7、14、21、28、35 d检测外周血细胞计数,在照射38 d检测血清SOD和GSH-PX活力、MDA含量,血清胆固醇和甘油三酯含量,肝脏脂代谢相关基因CYP7A1、FABP1、PPARα mRNA表达变化,计算主要脏器指数。结果 与模型对照组比较,在照射第35 d,SNPS中剂量组外周血血小板显著回升,在照射第38 d,SNPS各剂量组血清GSH-PX活力显著上升,血清MDA含量显著下降,肝脏中胆固醇7- 羟化酶含量显著回升。结论 SNPS对长期低剂量中子辐射损伤大鼠具有升高血小板,提高抗氧化能力,调节胆固醇代谢等保护作用。 |

Abstract: Objective To study the protective effects of Sipunculus nudus polysaccharides on radiation injury induced by low-dose neutron in rats. Methods Totally 50 male SD rats (sanitary degree,180-200 g) were administrated by intragastric infusion with SNPS at doses of 70, 140, 280 mg kg-1 before irradiation once a day for 38 days. Except for rats in the normal group, those in the rest groups were irradiated 22 h per day by 252Cf, the absorbed dose rate for 0.085 mGy h-1, integral absorbed dose for 71.06 mGy. At 7, 14, 21, 28, 35 d exposed irradiation, peripheral blood cells were counted. At 38 d exposed irradiation , the contents of malondialdehyde (MDA), the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX), total cholesterol (TC) and triglyceride (TG) in serum were detected. The genes expressions of CYP7A1 , FABP1 and PPARα in liver were analyzed. The indexes of main organs were calculated. Results Compared with model control group, peripheral blood PLT increased significantly in SNPSM group at 35 d exposed irradiation , GSH-PX activity and CYP7A1 (cholesterol 7-α-hydroxylase) gene expression increased significantly in SNPS groups, while the content of MDA decreased in SNPS groups at 38 d exposed irradiation . Conclusion SNPS may take a protective effect on rats injured by low-dose neutron irradiation with increasing PLT in serum, improving antioxidant activity and regulating cholesterol metabolism. |
