本文以大鼠为动物模型研究了海参皂苷单体Echinoside A(EA)在体内的消化吸收特性。Wistar大鼠适应饲养1周后,按体重分为10组,每组3只。禁食不禁水12h后,给大鼠分别按照30mg/kg?BW灌胃配置好的海参皂苷EA溶液1mL,分别在灌胃后0、0.5、1、2、3、4、7、9、11、24hr取血,建立并优化血清中海参皂苷EA的HPLC-MS检测方法,检测不同时间节点大鼠血中EA含量。实验结果显示,海参皂苷单体EA液质检测的定量限为3ng,0.3-20μg/mL浓度范围线性良好(R2=0.9946)。海参皂苷EA灌胃后在血中迅速出现,在3hr出现最高峰值,浓度达到0.83μg/mL,之后在7hr出现另一个较小峰,9hr后恢复至4hr水平(约0.24μg/mL),直至24hr。本文建立了血清中海参皂苷EA的液相色谱-质谱联用检测方法,发现海参皂苷单体EA可经消化道吸收,该结果为海参皂苷在保健食品及药物中的应用提供了科学依据。 |

To research the digestion and absorption characteristics of sea cucumber saponins Echinoside A (EA) in rats. Wistar rats were divided into 10 groups (n=3)according to the body weight. After acclimating for 1 week, the animals were fasted for 12 hours, and oral administration of 1 mL sea cucumber saponins EA solution at the dose of 30 mg/kg BW. The blood was collected in 0, 0.5, 1, 2, 3, 4, 7, 9, 11, 24 hr for the serum determination of EA . the HPLC-MS methods of serum EA determination was Established and optimized. The lower limit of quantification (LLOQ) of EA detected by HPLC-MS is 3 ng, EA showed a good linearity in the concentration range from 0.3 to 20 μg/mL (R2=0.9946). Sea cucumber saponins EA was appeared rapidly after oral administration, the highest peak concentration reached 0.83 μg/mL at 3 hr, another smaller peak showed at 7 hr, then return to the level of 4 hr(abut0.24μg/mL) at 9 hr and stay the level to 24hr. This paper established HPLC-MS method of sea cucumber saponins EA in serum, and found that the sea cucumber saponins EA could be absorpted by gastrointestinal, the results provided scientific evidence for the application of sea cucumber saponins in marine functional food and drugs. |
