      目的 观察羧甲基壳聚糖(CMCTS)和羟乙基壳聚糖(HECTS) 缓解大鼠干眼症状的效果。方法 分别以CMCTS和HECTS为主要原料配制了人工泪液,以SD大鼠为实验动物,通过缝合眼睑制作干眼动物模型,并借助Schirmer I试验、泪膜破裂时间检测、病理组织切片以及泪液成分分析等对人工泪液缓解大鼠干眼症状的效果进行评价。结果 相对于模型组,应用CMCTS滴眼液和HECTS滴眼液同透明质酸钠滴眼液(阳性对照组)一样,可显著促进大鼠受损眼表角结膜组织修复,穹窿部结膜杯状细胞数量大幅回升;滴眼7天后,泪膜稳定性及泪液分泌量即恢复至正常组的65%和85%,明显优于泪液缓冲液组。泪液初步分析结果表明CMCTS和HECTS可促进干眼症大鼠泪液中胃三乙酰甘油脂酶恢复正常。结论 CMCTS和HECTS可通过修复结膜杯状细胞明显缓解大鼠干眼症状。
English Summary:
      Objective To observe the mitigation effect of carboxymethyl chitosan (CMCTS) and hydroxyethyl chitosan (HECTS) on rat dry eye. Methods Artificial tears were prepared with CMCTS andHECTS as the main material, respectively. Rat model of dry eye was established by suturing the upper and lower eyelids of rats. The mitigation effect of artificial tears on rat dry eye was evaluated by Schirmer I test, BUT, histological observation. Results CMCTS and HECTS, similar to hyaluronate sodium (positive group) could promote the repair of damaged keratoconjuctivity, increase obviously the number of goblet cells in fornix as compared with the experimental group. After 7 days of the application of CMCTS and HECTS, the BUT and Shirmer values restored to 65% and 85% of the normal group, respectively, much better than those of the tear buffer group. Primary results from tear analysis revealed that CMCTS and HECTS could make the gastric triacylglycerol lipase back to normal. Conclusion CMCTS and HECTS could relieve the symptoms of dry eyes in rats by promoting the repairment of conjunctival goblet cells.
