目的 研究低聚古罗糖醛酸磷酸酯对RAW264.7细胞的激活作用。方法 低聚褐藻胶依次通过pH分级、高压酸解纯化的方法制备低聚古罗糖醛酸,进而采用磷酸/尿素法对其进行磷酸化修饰,制备低聚古罗糖醛酸磷酸酯。在低聚古罗糖醛酸磷酸酯对RAW264.7细胞激活实验中,分别测定了对RAW264.7细胞增殖、吞噬能力、细胞因子(TNF-α、IL-1β)释放、胞内酶(酸性磷酸酶、乳酸脱氢酶)活力的影响。结果 低聚古罗糖醛酸磷酸酯能显著促进RAW264.7细胞的增殖,提高其对中性红的吞噬,促进细胞因子的释放以及增强胞内酶的活性。结论 低聚古罗糖醛酸磷酸酯能够多层次、多水平的激活RAW264.7细胞,是一种潜在的免疫增强剂。 |

Objective To investigate the immunostimulatory effect of low-molecular-weight polyguluronate phosphate on RAW264.7 cell. Methods Low-molecular-weight polyguluronate (LPG) was obtained after the pH fractionation and acid hydrolysis of low-molecular-weight alginate, and then phosphorylated using H3PO4/Urea method to afford low-molecular-weight polyguluronate phosphate (LPGP). In the immunostimulatory assay, cells proliferation, phagocytosis, cytokines (TNF-α, IL-1β) release and activities of intracellular enzymes (acid phosphatase, lactate dehydrogenase) were determined to study the effects of LPGP on RAW264.7 cell. Results LPGP could significantly improve the cell proliferation, the neutral red phagocytosis, cytokines release and intracellular enzymes activities of RAW264.7 cell. Conclusion LPGP could effectively activate RAW264.7 cell via multiple levels. It indicated that LPGP was a potential immunoenhancer. |
