目的 研究藻酸双脂钠(PSS)联合更昔洛韦(GCV)在体外抗人巨细胞病毒(HCMV)感染的作用。方法 应用体外细胞培养技术构建HCMV-AD169 感染人胚肺成纤维细胞(HELF)模型,对照组采用GCV (50 mg.L-1)单一药物,实验组采用PSS (100 mg.L-1)和GCV (50 mg.L-1)的联合药物,应用两组药物同时对模型进行干预,观察干预后两组的特征性致细胞病变效应(CPE)的变化,同时应用RT-PCR法检测用药后两组HCMV的IE2的表达情况。 结果 实验组和对照组均可提高感染细胞的存活率,同时降低感染细胞中HCMV IE2的mRNA表达量。显微镜下形态学观察显示,相比对照组,实验组能明显降低药物对HELF细胞的毒性作用,提高细胞存活率。结论 PSS联合GCV具有良好的体外抗HCMV作用,二者具有协同抗病毒作用。 |

Objective To study the effect of ganciclovir (GCV) combined with propylene glycol alginate sodium sulfate (PSS) on human cytomegalovirus (HCMV) infection in vitro. Methods A human embryonic lung fibroblast (HELF) model of HCMV-AD169 infection was established in vitro. GCV (50 mg.L-1) alone or in combination with PSS (100 mg.L-1) was added to HCMV-infected HELF cultures and the characteristic cytopathic effects were assessed. RT-PCR was used to evaluate HCMV IE2 (Immediate-Early 2) mRNA expression. Results Both GCV and GCV plus PSS increased the survival rate and decreased the expression of HCMV IE2 mRNA in infected HELFs. The morphological changes observed by light microscopy showed that the survival rate was increased and the toxic effect on the HELFs was weakened in GCV plus PSS group. Conclusions PSS combined with GCV has a good anti-HCMV effect in vitro, and these drugs have synergistic antiviral effects. |
