      目的 探究脱脂鳕鱼卵对生长期小鼠长骨生长的促进作用及相关机制。方法 采用3周龄初断乳ICR雄性小鼠为生长期动物模型,连续饲喂脱脂鳕鱼卵21d后,分别检测小鼠体重、身长、尾长、骨生长速率和血清骨生成指标的变化;观察小鼠股骨生长板组织形态学变化;检测小鼠肝脏、股骨生长板GH/IGF-1信号轴相关基因表达的变化。结果 脱脂鳕鱼卵能显著促进生长期小鼠身长、尾长、股骨和胫骨等纵向骨生长,但对体重无显著影响;显著提升小鼠骨生长速率;显著增加血清GH(P<0.05)、IGF-1(P<0.01)、BMP-2(P<0.01)和ALP(P<0.01)的含量;显著提高股骨生长板整体高度(P<0.01)、增殖区高度(P<0.01)与肥大区高度(P<0.01);显著促进肝脏、股骨GH-IGF-1信号轴相关基因的表达。结论 脱脂鳕鱼卵能通过调控生长期小鼠体内GH/IGF-1信号轴相关基因的表达来促进骨生成和软骨内成骨,最终促进长骨生长。
English Summary:
      Objective To evaluate the ameliorating of defatted cod eggs on longitudinal bone growth in adolescent mice and explore the related mechanism. Methods Using 3-week-old ICR male mice as the model of growing animals, the mice were continuously fed with defatted cod eggs for 21 days. After 21 days treatment, the weight, body length and tail length of mice were determined. Tetracycline hydrochloride was injected intraperitoneally to form fluorescent bands on the growth plates for measuring the longitudinal bone growth rate. The effects of defatted cod eggs on the femoral growth plate structure and GH-IGF-1 pathway related genes mRNA expression in mice were tested. Results Defatted cod eggs could respectively markedly increase the body length and tail length of adolescent mice; promote the development of femur and tibia; cause a rise of GH, IGF-1, BMP-2 and ALP levels in serum. There was a significant increase in the rate of bone growth in the defatted cod eggs group compared to the control group. The results showed that defatted cod eggs could significantly increase the overall height of growth plate, the length of proliferous zone and hypertrophic zone, as well as the expression of GH-IGF-1 pathway related genes in liver and femur from the transcriptional level. Conclusion Defatted cod eggscould promote the development of longitudinal bone growth of adolescent mice by regulating the GH / IGF-1 pathway in vivo.
