目的 对来源于南极海洋丝状真菌Lecanicillium kalimantanense HDN13-339产生的胞外多糖的结构和抗氧化活性进行研究。方法 利用乙醇沉淀法、强阴离子交换色谱柱和凝胶渗透色谱柱对菌株所产胞外多糖进行分离纯化;通过PMP柱前衍生高效液相色谱法、红外光谱、气质联用色谱和核磁共振波谱对多糖的结构进行表征;通过测定DPPH自由基、超氧阴离子自由基、羟基自由基、ABTS自由基清除活性及Fe2+螯合能力研究胞外多糖的抗氧化活性。结果 从南极海洋丝状真菌Lecanicillium kalimantanense HDN13-339发酵产物中分离得到多糖HDN-51,其分子量为16.3 kDa,主要由甘露糖和半乳糖构成,糖链是由(1→)-Galf、(1→2)-Manp、(1→5)-Galf、(1→6)-Manp、(1→6)-Galf和(1→2,6)-Manp组成,HDN-51具有良好的抗氧化活性,特别是羟基自由基清除能力。结论 首次从南极海洋丝状真菌Lecanicillium kalimantanense HDN13-339中分离得到一种结构新颖的半乳甘露聚糖HDN-51,其具有一定的抗氧化活性。 |

Objective To study the structural characterization and antioxidant activity of the exopolysaccharide from the Antarctic marine filamentous fungus Lecanicillium kalimantanense HDN13-339. Methods An exopolysaccharide was isolated from the culture medium of the Antarctic marine filamentous fungus Lecanicillium kalimantanense HDN13-339 using ethanol precipitation, anion-exchange and gel-permeation chromatography. The structural characteristics of the exopolysaccharide were identified by PMP-HPLC, IR, GC-MS and NMR. The antioxidant activity of the exopolysaccharide was evaluated with the scavenging abilities on DPPH residues, superoxide residues, hydroxyl residues, ABTS residues and Fe2+ chelating property. Results The polysaccharide HDN-51 was obtained from the Antarctic marine filamentous fungus Lecanicillium kalimantanense HDN13-339, and its average molecular weight was estimated to be about 16.3 kDa. HDN-51 was mainly composed of mannose and galactose. The chain of HDN-51 mainly consisted of (1→)-Galf, (1→2)-Manp, (1→5)-Galf, (1→6)-Manp, (1→6)-Galf and (1→2,6)-Manp. HDN-51 had a high antioxidant activity, especially in the scavenging ability on hydroxyl residues. Conclusion HDN-51 was firstly obtained from the Antarctic marine filamentous fungus Lecanicillium kalimantanense HDN13-339, and it was a novel galactomannan with antioxidant ability. |
