      目的 对从青岛胶东海岸潮间带淤泥中分离筛选高产胞外硫酸软骨素酶的菌株进行鉴定和发酵条件优化。方法 通过平板快速筛选法和摇瓶复筛,筛选到1株高产胞外硫酸软骨素裂解酶的菌株C26,根据形态学和16S rRNA基因序列分析对其鉴定,并采用单因素实验和响应面实验对发酵条件进行优化。结果 根据形态学和16S rRNA基因序列分析,鉴定菌株C26为不动杆菌属(Acinetobacter sp.),命名为Acinetobacter sp. C26。优化最适发酵条件为:硫酸软骨素15 g?L-1,酵母粉7.69 g?L-1,培养温度23 ℃,在最适条件下硫酸软骨素酶的活力可达11.96 U/mL,是优化前的4.82倍。结论 本研究筛选得到的菌株Acinetobacter sp. C26产酶活力高,发酵周期短,在生物化工及医药方面具有潜在应用价值。
English Summary:
      Objective To screen Strains with high-yield extracellular chondroitinase from the intertidal mud of Qingdao Jiaodong coast,identify the high-yield strain and optimize the fermentation conditions. Methods The strain C26 was screened by plate method for rapid screening and shake flask for re-screening. The strain C26 was identified by morphological characteristics and the analysis of 16S rRNA. The fermentation conditions of C26 were optimized by single factor experiment and response surface experiment. Results The strain C26 was identified as Acinetobacter sp. according to its morphological characteristics and analysis of 16S rRNA, which was named as Acinetobacter sp. C26. The optimum fermentation conditions were as follows:chondroitin sulfate 15 g?L-1, yeast powder 7.69 g?L-1, culture temperature23 ℃. The activity of chondroitinase could reach to 11.96 U/mL in the optimum culture conditions, which was 4.82 times than before. Conclusion The results showed that the strain Acinetobacter sp. C26 had high activity and short fermentation cycle, which might have potential application value in biochemical and medicine.
