      目的 探讨仿刺参糖胺聚糖(HGAG)对肺结核和肺癌患者免疫调节功能影响。方法 选取健康人群40名(健康组),肺结核患者30例(结核组)和肺癌患者30例(肺癌组),取抗凝外周血4 mL,采用Ficoll密度梯度离心法分离PBMC,体外与不同浓度HGAG共培养24 h。运用流式细胞仪检测CD2、CD28和B7分子的表达。结果 健康组CD2、CD28和B7-2表达在10μg·mL-1最明显,而B7-1以50μg·mL-1最明显,与对照组比较均具有差异性(P<0.05);肺结核组CD2、CD28和B7-1表达在50μg·mL-1最明显,B7-2以10μg·mL-1表达最明显,与对照组比较均具有差异性(P<0.05);肺癌组CD2、CD28,B7(B7-1和B7-2)在100μg·mL-1时表达最明显,与对照组比较均具有差异性(P<0.05)。结论 HGAG在一定浓度范围内可以不同程度促进CD2、CD28和B7分子表达,体外调节肺结核和肺癌患者的细胞免疫功能。
English Summary:
      Objectives Study the effect of Apostichopus japonicus glycosaminoglycan (HGAG) on immune regulation in patients with pulmonary tuberculosis and lung cancer. Methods 40 healthy people (healthy group), 30 pulmonary tuberculosis patients (tuberculosis group) and 30 lung cancer patients (lung cancer group) were selected and anticoagulation Peripheral blood 4mL were taken. The peripheral blood mononuclear cells (PBMC) were isolated by Ficoll density gradient centrifugation, and cultured in vitro with different concentrations of HGAG for 24 h. The expressions of CD2, CD28 and B7 molecules were detected by flow cytometry respectively. Results The expression levels of CD2, CD28 and B7-2 in healthy group were the most obvious at 10μg·mL-1, while the expression levels of B7-1 were the most obvious at 50μg·mL-1, which were both different from those in control group (P<0.01). The expression of CD2, CD28 and B7-1 in tuberculosis group were the most obvious at 50μg·mL-1, while the expression levels of B7-2 were the most obvious at 10μg·mL-1, which were both different from those in control group (P<0.01). The expression of CD2, CD28 and B7 (B7-1 and B7-2) in lung cancer group was the most obvious at 100μg·mL-1, which was different from that in control group (P<0.05). Conclusion HGAG can promote the expression of CD2, CD28 and B7 molecules in varying degrees, regulating the cellular immune function of pulmonary tuberculosis and lung cancer patients in vitro.
