目的 对耐酸产紫青霉OUCMDZ-019的次级代谢产物进行化学成分和生物活性研究,发现潜力结构资源。方法 对菌株进行规模发酵得到粗提物,利用常压柱层析、凝胶分子筛以及半制备高效液相等方法进行次级代谢产物的分离纯化;利用现代波谱学方法(核磁共振1D NMR、2D NMR、圆二色谱ECD、旋光等)对化合物进行结构鉴定,并评价其抗病毒生物活性。结果 分离鉴定得到4个单体化合物,(-)-Mitorubrinol (1)、6″-Hydroxy-(R)-mitorubrinic acid (2)、(-)-Mitorubrinic acid (3)、Purpurquinone D (4)。结论 从耐酸真菌产紫青霉OUCMDZ-019的次级代谢产物中分离鉴定得到了化合物1~4,均属于嗜氮酮类化合物,研究获得规模量产化合物3,为后续药物的结构衍生打下基础。 |

Objective To study the secondary metabolites of acid resistant fungus Penicillium purpureum OUCMDZ-019. Methods The crude extracts were obtained by large-scale fermentation. The secondary metabolites were separated by column chromatography, gel molecular sieve and semi high performance liquid chromatography. The structures of the compounds were identified by modern spectroscopy methods (nuclear magnetic resonance 1D NMR, 2D NMR, circular dichroism ECD, optical rotation, etc.). Results Four compounds were isolated and identified as (-)-mitorubinol (1), 6″-hydro-(R)-mitorubinic acid (2), (-)-mitorubinic acid (3) and purpurquinone D (4). Conclusion Compounds 1~4 were isolated and identified from the secondary metabolites of acid resistant fungus Penicillium purpureum OUCMDZ-019, which belong to azaphilones. The large-scale mass production of compound 3 was obtained, which made a base for the structural derivation of subsequent drugs. Keywords: Acid tolerant fungi; Azaphilones; Penicillium |
