目的 建立柱前衍生高效液相色谱法(HPLC)法测定紫贝齿饮片中11种氨基酸的方法,并比较不同基原的差异。方法 以异硫氰酸苯酯为柱前衍生化试剂,采用C18色谱柱,流动相A为乙腈,流动相B为0.1mol·L-1醋酸钠溶液(pH 6.5),梯度洗脱,检测波长254 nm,色谱柱温度40℃。运用主成分分析(PCA)、偏最小二乘法判别分析(PLS-DA)和方差分析研究了紫贝齿中的4种基原的氨基酸成分的差异。结果 11种氨基酸呈良好的线性关系(R>0.999);精密度、稳定性、重复性试验RSD值分别小于1.77%、2.54%、1.98%;平均回收率在90.7%~108.9%(RSD为0.67%~1.58%,n = 6)之间。紫贝齿4种基原之间氨基酸成分存在差异,并发现3种氨基酸差异性成分。结论 建立的氨基酸测定方法稳定可靠,可用于紫贝齿饮片的质量评价。 |

Objective To establish a new pre-column derived HPLC method to quantify the levels of 11 amino acids in Conche Cypeaeae Violacae, and compare the differences between different origins. Methods Hydrolytic amino acids were derivatized with phenyl isothiocyanate and the samples were analyzed on a C18 column with gradient elution of acetonitrile (A) and 0.1 mol·L-1 sodium acetate solution (pH 6.5) (B). The detected wavelength was 254 nm, the chromatographic column temperature was set at 40℃. The differences in amino acid composition of four types in Conche Cypeaeae Violacae were studied by principal component analysis (PCA), partial least squares discriminant analysis (PLS-DA), and analysis of variance. Results 11 amino acids showed a good linear relationship (R>0.999). The RSD values of density, stability, and repeatability tests were less than 1.77%, 2.54%, and 1.98%, respectively. The average recovery rate ranges from 90.7% to 108.9%(RSD 0.67%~1.58%,n = 6). There were differences in amino acid composition among the four types of Conche Cypeaeae Violacae, and three different amino acid components were found. Conclusion The established amino acid determination method was stable and reliable, and can be used for the quality evaluation of Conche Cypeaeae Violacae. |
