目的 研究κ-卡拉胶寡糖在大鼠体内的药代动力学特征。方法 建立并验证了聚合度为3、5、7的κ-卡拉胶寡糖在大鼠体内的高效液相色谱-质谱联用(LC-MS/MS)检测方法。大鼠灌胃和尾静脉注射给予不同聚合度的κ-卡拉胶寡糖后,收集不同时间点的血浆样品。测定血浆样品中的κ-卡拉胶寡糖浓度计算生物利用度等药代动力学参数,考察不同聚合度κ-卡拉胶寡糖在大鼠体内的药代动力学特征。结果 大鼠尾静脉注射剂量为4 mg/kg 的κ-3、κ-5和κ-7卡拉胶寡糖后,达峰时间为2 min,达峰浓度分别为5.42、1.96、0.524 μM,大鼠灌胃剂量为40 mg/kg 的κ-3、κ-5和κ-7卡拉胶寡糖后,κ-3的达峰时间为60min,达峰浓度为0.085 μM,κ-5、κ-7卡拉胶寡糖灌胃后,不同时间点检测得到的血浆中药物浓度很低。结论 κ-卡拉胶寡糖的吸收程度随着聚合度的增加而降低。 |

Objective To investigate the pharmacokinetic characteristics of κ-carrageenan oligosaccharides in rats. Methods A high-performance liquid chromatography-mass spectrometry (LC-MS/MS) detection method for κ-carrageenan oligosaccharides with a degree of polymerization of 3, 5, and 7 in rats was established and verified. Plasma samples were collected at different time points after intragastric administration and tail vein injection of κ-carrageenan oligosaccharides with different degrees of polymerization. The concentration of κ-carrageenan oligosaccharides in plasma samples was measured to calculate the bioavailability and other pharmacokinetic parameters, and the pharmacokinetic characteristics of κ-carrageenan oligosaccharides with different degrees of polymerization in rats were investigated. Results The peak time of κ-3, κ-5 and κ-7 carragenan oligosaccharides was 2 min and the peak concentrations were 5.42, 1.96 and 0.524 μM, respectively, after the tail vein was injected with 4 mg/kg of κ-3, κ-5 and κ-7 carragenan oligosaccharides by intragastric administration. The peak time of κ-3 was 60 min and the peak concentration was 0.085 μM, while the plasma concentration of κ-5 and κ-7 carrageenan oligosaccharides was very low at different time points after intragastric administration of 40 mg/kg of κ-3, κ-5 and κ-7 carrageenan oligosaccharides to rats. Conclusion The absorption degree of κ-carrageenan oligosaccharides decreased with the increase of polymerization degree. |
