      目的 评价脱细胞鱼皮敷料的性能和促愈合作用,为该敷料的进一步开发应用奠定基础。方法 将罗非鱼鱼皮经脱细胞、除杂、病毒灭活、冻干等处理后,获得脱细胞鱼皮敷料(AFSD)。通过扫描电子显微镜和透射电子显微镜观察AFSD的结构特征。通过测定孔隙率和液体吸收性评价敷料的吸液能力。通过测定外源性DNA含量、内毒素含量、组织成分染色、细胞毒性试验初步评估AFSD的生物相容性。并建立SD大鼠背部全层皮肤缺损模型,分别在术后不同时间点计算创面愈合率,评价AFSD的促愈合作用;取创面标本进行组织病理观察,分析血管生成、炎症反应和组织再生情况。结果 AFSD一面疏松一面致密,主要成分为胶原,孔隙率为(65.56±3.0)% ,液体吸收倍数为(7.722±0.14)倍,DNA残留量为(12.21±3.3)ng/mg干重,细菌内毒素含量为(0.4155±0.018)EU/mL,细胞毒性试验中100%浸提液24 h的细胞存活率为(107.3±2.4)%。在大鼠急性全层皮肤缺损创面促进愈合实验中,有明显的促进作用,术后28天时,相较于对照组的表皮层更完整、炎症反应更低、瘢痕化不明显。结论 AFSD生物相容性优良,具有明显的促进愈合作用,能够改善创后修复瘢痕问题,有望开发成新型生物敷料。
English Summary:
      Objective To evaluate the property and wound healing effect of acellular fish skin dressing, and lay a foundation for its further development and application. Methods Acellular fish skin dressing(AFSD) was obtained after tilapia skin was decellularized, impurities removed, virus inactivated, and freeze-dried. The structural characteristics of AFSD was observed by scanning electron microscopy and transmission electron microscopy. The fluid absorption capacity of AFSD was evaluated by measuring porosity and fluid absorbency. The biocompatibility of AFSD was initially evaluated by measuring exogenous DNA content, endotoxin content, tissue component staining, and cytotoxicity test.We also established a full-thickness skin defect model on the back of SD rats, calculated the wound healing rate at different time points after surgery, and evaluated the healing effect of AFSD. Wound specimens were taken for histopathological observation, analyze angiogenesis, inflammatory response and tissue regeneration. Results AFSD was loose on one side and dense on the other, the main component was collagen, the porosity was (65.56±3.0)%, the liquid absorption multiple was (7.722±0.14) times, and the DNA residual amount was (12.21±3.3) ng/mg dry weigh, the bacterial endotoxin content was (0.4155±0.018) EU/mL, the cell survival rate of 100% extract solution for 24 hours in the cytotoxicity test was (107.3±2.4)%. In the experiment on promoting the healing of acute full-thickness skin defect wounds in rats, it had a significant promoting effect. Compared with the control group, the epidermis layer was more complete, the inflammatory response was lower, and the scarring was less obvious at 28 days after surgery. Conclusion AFSD had excellent biocompatibility and obvious healing-promoting effects, it could improve the post-traumatic repair of scarring problems, and was expected to be developed into a new type of biological dressing.
