目的 对来源于泰国红树林泥土样品的放线菌Nocardiopsis sp. OUCMDZ-4937进行菌种鉴定及次级代谢产物的研究。方法 对菌株进行规模发酵得到提取物,利用柱色谱以及半制备高效液相色谱(HPLC)等手段进行次级代谢产物的分离纯化;通过核磁共振(NMR)、质谱(MS)等现代波谱学方法并结合文献对所得化合物进行结构鉴定,测试了化合物对人体致病菌的抑制活性。结果 分离鉴定出2个新化合物,包含1个腺嘌呤衍生物2-thiomethyl-3-methyl zeatin(1)和1个芳基丁内酯类化合物noreucol B(2)。生物活性测试结果显示,化合物2对枯草芽孢杆菌有微弱的抑制作用。结论 从放线菌Nocardiopsis sp. OUCMDZ-4937的次级代谢产物中分离鉴定了2个新化合物,为后续生物活性研究奠定了一定的化合物基础。 |
English Summary: |
Objective To study the secondary metabolites of Nocardiopsis sp. OUCMDZ-4937, an actinomycete isolated from a mangrove mud sample collected from Thailand. Methods The crude extracts were obtained by large-scale fermentation. The secondary metabolites were separated and purified by silica gel column chromatography, thin layer chromatography and semi-HPLC column chromatography, and their structures were determined by NMR, MS, ECD and other spectral methods. The inhibitory activity of the compounds against human pathogenic bacteria was assayed. Results Two new compounds were isolated from the secondary metabolites of Nocardiopsis sp. OUCMDZ-4937, including one adenine derivative 2-thiomethyl-3-methyl zeatin (1) and one arylbutyrolactone compound noreucol B (2). The results of biological activity test showed that compound 2 had a weak inhibitory effect on Bacillus subtilis. Conclusion Compounds 1 and 2 were isolated and identified from the secondary metabolites of an actinomycete strain isolated from mangrove mud samples. This study laid the foundation for subsequent biological activity research. |
