| 目的 针对创面出血及后续由继发感染导致的愈合缓慢等问题,制备兼具吸水锁水和抗菌功能的淀粉/羧甲基壳聚糖-聚赖氨酸微球(St/CS-EPLM3)并研究其止血、促愈合性能,为新型复合止血微球开发提供新策略。方法 通过反相乳液聚合法制备微球,分别以大出血(小鼠断尾、股动脉破裂实验)和小出血模型(肝损伤实验)评估其止血效果,以背部全皮肤损伤模型评价其促愈合效果。结果 St/CS-EPLM3表现出高吸水率(588%)和高孔隙率(52.8%)。小鼠断尾实验中St/CS-EPLM3组平均出血量为115 mg,止血时间为88 s;股动脉破裂实验中,其平均出血量为417 mg,止血时间为93 s;在肝损伤实验中,其平均出血量为45 mg,止血时间为52 s。此外,St/CS-EPLM3组对金黄色葡萄球菌和大肠杆菌均表现出最佳抗菌效果,皮肤创面愈合率在7天内达89.8%,H&E和Masson染色提示创面炎症降低明显、胶原再生显著。结论 St/CS-EPLM3能显著降低多种创面出血模型的出血量与出血时间从而快速止血,其中对大出血的止血效果优于小出血,同时有效预防继发感染并加速愈合,与已上市止血粉相比优势明显,具有较大应用潜力。 |

| Objective To solve the problems of wound bleeding and subsequent slow healing caused by secondary infections, starch/carboxymethyl chitosan-polylysine microspheres (St/CS-EPLM3) with both water-absorbing function, water-locking function and antimicrobial function were prepared, and their hemostatic and healing properties were investigated, which provided a new strategy for the development of new composite hemostatic microspheres. Methods Microspheres were prepared by reversed-phase emulsion polymerization, and their hemostatic effect was assessed by major hemorrhage(caudal hemorrhage test and femoral artery hemorrhage test in mice)and minor hemorrhage(liver injury test in mice), and their pro-healing properties were evaluated by establishing a total skin injury on the back. Results St/CS-EPLM3 exhibited high water absorption ratio (588%) and high porosity ratio (52.8%). The average amount of bleeding was 115 mg and hemostasis time was 88 s in caudal hemorrhage test; The average amount of bleeding was 417 mg and hemostasis time was 93 s in femoral artery hemorrhage test; The average amount of bleeding was 45 mg and hemostasis time was 52 s in liver injury test.Moreover, St/CS-EPLM3 showed a positive effect on both S.aureus and E.coli and the healing rate of the skin wounds could reach 89.8% within 7 days, and the results of H E and Masson staining demonstrated that St/CS-EPLM3 led to a significant reduction of wound inflammation and significant collagen regeneration. Conclusion St/CS-EPLM3 significantly reduced the amount of bleeding and bleeding time in a variety of hemorrhage models,exhabiting better hemostatic effect compare with minor hemorrhage, and prevented the wound from developing secondary infections and accelerated the wound healing rate, which had obvious advantages over commercially available hemostatic powders, suggesting that St/CS-EPLM3 may have a greater potential. |
