目的 对来源于中国西沙群岛海绵(Acanthella cavernosa)共附生真菌Aspergillus flocculosus(22-11-ZP-2)的次级代谢产物进行研究。方法 利用硅胶柱层析,薄层色谱法,高效液相色谱法,中压制备液相色谱法等方法进行次级代谢产物的分离和纯化,利用质谱,核磁共振波谱等现代谱学方法结合相关文献数据分析对单体化合物进行结构鉴定。结果 从真菌Aspergillus flocculosus分离鉴定得到10个聚酮类化合物,分别为asperlactone (1)、aspilactonol B (2)、aspilactonol C (3)、aspilactonol D (4)、asperochralactone C (5)、dihydropenicillic acid (6)、asperochratide G (7)、8,9-dihydroxy-8,9-deoxyaspyrone (8)、dihydroaspyrone (9)、5-Methoxy-8,9-dihydroxy-8,9-deoxyaspyrone (10)。活性测试结果表明:化合物4、8 对斑马鱼具有尾静脉血栓预防活性,浓度为20 μmol/L;化合物3、5、8具有明显的促血管生成活性;化合物1、6、9具有微弱的促血管生成活性。 |

Abstract: Objective To study the secondary metabolites of the sponge (Acanthella cavernosa)-associated fungus Aspergillus flocculosus (22-11-ZP-2) from the Xisha Islands, China. Methods Silica gel column chromatography, thin layer chromatography, high-performance liquid chromatography, and medium-pressure preparative liquid chromatography were employed for the separation and purification of metabolites. Modern spectroscopy methods such as mass spectrometry and nuclear magnetic resonance spectroscopy combined with relevant literature data analysis were used to identify the structures of monomers. Results Ten sesquiterpenoids were isolated from Aspergillus flocculosus and were identified as asperlactone (1)、aspilactonol B (2)、aspilactonol C (3)、aspilactonol D (4)、asperochralactone C (5)、dihydropenicillic acid (6)、asperochratide G (7)、8,9-dihydroxy-8,9-deoxyaspyrone (8)、dihydroaspyrone (9)、5-Methoxy-8,9-dihydroxy-8,9-deoxyaspyrone (10). The results of the activity tests showed that compounds 4 and 8 had preventive activity against caudal vein thrombosis in zebrafish at a concentration of 20 μmol/L. Compounds 3, 5 and 8 exhibited significant pro-angiogenic activity, while compounds 1, 6 and 9 had weak pro-angiogenic activity. |
