| 目的 基于深海链霉菌-多重耐药菌(Multidrug-resistant bacteria, MDR bacteria)双层平板共培养的筛选方法,定位具有抗MDR菌活性的深海链霉菌,并挖掘其活性次级代谢产物,为海洋天然产物开发提供理论参考。方法 采用双层平板进行共培养确定抗菌活性菌株。采用OSMAC(One Strain Many Compounds)策略优化发酵条件,积累次级代谢产物。通过中压色谱柱(MPLC)、HPLC等手段进行化合物分离纯化,解析化合物结构并评估其对MDR菌的最小抑菌浓度。结果 从海洋菌株库中筛选得到次级代谢产物具有抗MDR菌活性的深海链霉菌Streptomyces sp. OUCT16-37,并从F2培养基的粗提物中分离得到两个16元多烯大环内酯类化合物Elaiophylin(1)和11,11'-O-Dimethylelaiophylin(2)。Elaiophylin(1)对多重耐药S. aureus CCARM 3090、E. faecalis CCARM 5172、E. faecium CCARM 5203表现出中等抑菌活性。结论 基于深海链霉菌-多重耐药菌双层平板共培养的方法筛选到抗菌活性候选菌株OUCT16-37,从中挖掘到一类具有抗MDR菌活性的Elaiophylin类多烯大环内酯(1和2),为发掘抗菌活性菌株提供了简便高效的筛选方法,为深海微生物的资源开发奠定基础。 |

| Objective A screening method based on deep-sea Streptomyces-MDR bacteria (Multidrug-resistant Bacteria) double-layer plate co-culture for identifying anti-MDR active deep-sea Streptomyces and mining their bioactive secondary metabolites, providing theoretical reference for marine natural product development. Methods A dual-layer plate co-culture assay was used to determine the antibacterial active strains. To enhance secondary metabolite production, the OSMAC (One Strain Many Compounds) approach was applied to optimize fermentation conditions. Target compounds were isolated and purified through MPLC and HPLC, and the structure of the compounds was analyzed and the minimum inhibitory concentration (MIC) against MDR bacteria was evaluated. Bioinformatics was used to predict the biosynthetic gene cluster in Streptomyces sp. OUCT16-37 genome and annotate the gene function. Results From a marine-derived strain library, Streptomyces sp. OUCT16-37 was identified as a producer of anti-MDR secondary metabolites. Bioactivity-guided fractionation of the F2 culture medium extract yielded two 16-membered polyene macrolides: elaiophylin (1) and 11,11'-O-dimethylelaiophylin (2). Elaiophylin (1) exhibited moderate antibacterial activity against MDR strains, including S. aureus CCARM 3090, E. faecalis CCARM 5172, and E. faecium CCARM 5203. Conclusion Based on the method of deep-sea Streptomyces-MDR bacteria double-layer plate co-culture, the candidate strain OUCT16-37 with antibacterial activity was screened, and a class of elaiophylin polyene macrolides (1 and 2) with anti-MDR activity were excavated from it, which provided a simple and efficient screening method for discovering antibacterial strains and laid a foundation for the resource development of deep-sea microorganisms. |
