      目的 对南海来源软珊瑚Clavularia sp.中化学成分多样性及其生物活性进行研究。方法 利用正相硅胶、反相十八烷基硅烷键合硅胶(ODS-C18)、凝胶Sephadex LH-20和半制备高效液相(semi-HPLC)等色谱学方法对软珊瑚乙酸乙酯提取物中的化学成分进行分离纯化;利用旋光仪、核磁共振波谱(NMR)、质谱(MS)等波谱学方法,结合单晶数据与文献的数据对比确定化合物的结构。结果 从软珊瑚的乙酸乙酯提取物中分离鉴定了14个海兔烷型二萜类化合物,分别为clavirolide K (1)、clavirolide J (2)、clavirolide O (3)、clavirolide R (4)、clavudiol A (5)、clavudiol B (6)、clavurol F (7)、clavirolide E (8)、clavirolide B (9)、clavulactone (10)、clavirolide C (11)、clavirolide D (12)、clavularinlide A (13)和clavularinlide B (14)。化合物5、3和6在10 μmol/L物质的量浓度下对脂多糖(LPS)诱导NO产生的抑制率分别为27.6%、27.1%和23.5%。结论 从南海来源软珊瑚Clavularia sp.的乙酸乙酯提取物中分离得到了化合物1~14,其中化合物5、3和6对LPS诱导NO产生具有一定抑制作用。
English Summary:
      Objective To study the chemical diversity and biological activity of Clavularia sp., a soft coral from the South China Sea. Methods The chemical components in the ethyl acetate extract of soft coral were separated and purified by normal silica gel, reverse octadecyl silane bonded silica gel (ODS-C18), gel Sephadex LH-20 and semi HPLC; using spectroscopic methods such as polarimeter, nuclear magnetic resonance spectroscopy (NMR), and mass spectrometry (MS), and comparing single crystal data with literature data to determine the structure of the compound. Results 14 dolabellane diterpenoid compounds were isolated and identified from the ethyl acetate extract of soft coral, including clavirolide K(1), clavirolide J(2), clavirolide O (3), clavirolide R(4), clavudiol A(5), clavudiol B(6), clavurol F(7), clavirolide E(8), clavirolide B(9), clavulactone(10), clavirolide C(11), clavirolide D(12), clavularinlide A(13), clavularinlide B(14). The inhibition rates of compounds 5, 3, and 6 on lipopolysaccharide (LPS) induced NO production at a concentration of 10 μmol/L were 27.6%, 27.1%, and 23.5%, respectively. Conclusion Compounds 1~14 were isolated from the ethyl acetate extract of Clavularia sp., a soft coral from the South China Sea. Compounds 5, 3, and 6 exhibited certain inhibitory effects on LPS induced NO production.
