      摘 要:目的 对饱和褐藻寡糖(Saturated alginate oligosaccharide, SAOS)开展系统分析,并探讨其对正常动物学习记忆能力的改善作用。方法 采用亚临界酸降解方法制备的SAOS,通过分子排阻色谱法(SEC)、质谱法(MS)、傅里叶变换红外光谱法(FT-IR)和核磁共振波谱法(NMR)对其相对分子量、聚合度及结构进行分析。将Balb/c雄性小鼠(用于跳台和避暗实验)和SD雄性大鼠(用于穿梭箱实验)分为阴性对照组和SAOS低(0.083 g/kg BW)、中(0.17 g/kg BW)、高(0.50 g/kg BW)剂量组,连续灌胃30天后,进行跳台、避暗和穿梭箱实验,每个实验中每组各10只老鼠。结果 SAOS的重均分子量(Mw)为880 Da,数均分子量(Mn)为740 Da,平均聚合度(DP)约为4,为饱和褐藻寡糖结构。在行为学实验中,SAOS能延长跳台实验小鼠的潜伏期,延长避暗实验小鼠的潜伏期、减少进入暗室的动物数和平均错误反应次数,降低穿梭箱实验大鼠的主动回避平均反应时间和被动回避平均反应时间。结论 SAOS具有提高正常动物学习记忆能力的作用。

      Abstract: Objective To conduct a systematic analysis of the saturated alginate oligosaccharide (SAOS), and investigate its effects on normal animal learning and memory abilities. Methods SAOS was prepared by subcritical acid degradation, and its relative molecular mass, degree of polymerization (DP) and structure were analyzed by size exclusion chromatography (SEC), mass spectrometry (MS), fourier transform infrared spectroscopy (FT-IR) and nuclear magnetic resonance spectroscopy (NMR). Balb/c male mice (for step down test and dark avoidance test) and SD male rats (for shuttle box test) were divided into four groups, including the negative control group, and the low-dose (0.0834 g/kg BW), the medium-dose (0.1667 g/kg BW) and the high-dose (0.5000 g/kg BW) groups of SAOS. After continuous gavage for 30 days, the animals were directly subjected to the step down test, dark avoidance test and shuttle box test, with 10 animals per group in each test. Results The weight-average molecular weight (Mw) and the number-average molecular weight (Mn) of SAOS are 880 Da and 740 Da, respectively. SAOS has a DP of around 4, and hold a saturated oligosaccharide structure. In behavioral test, SAOS can prolong the latency in the step down test and dark avoidance test, reduce number of animals entering the dark room and the average number of errors in the dark avoidance test, and decrease the average response time of active avoidance and passive avoidance in the shuttle box test. Conclusion SAOS can improve the learning and memory ability of normal animals.
